The doctor was bundled into a private car and rushed to a hospital.
Netcare911 spokesperson Shawn Herbst said paramedics understood that a group of armed robbers stormed into the surgery in Peter Magano Road and demanded cash.
Police are investigating a murder and two attempted murders, police spokesperson Captain Kay Makhubele said.
"While the motive is still unclear at this stage, it has, during the preliminary investigation, emerged that about four unknown male suspects, all wearing facial masks, arrived at the surgery in a Toyota Quantum, fitted with false registration number plates, and started firing shots indiscriminately," said Makhubele.
"On arrival at the scene, police found a woman who was declared dead by paramedics, and two other victims - a man and a woman - who were both injured and rushed to hospital for medical attention."
Makhubele added that police are not at liberty to disclose the identities of the victims at this stage of the investigation.
"A manhunt is under way in search of the suspects and police are appealing to anyone who may have witnessed the incident or have information that could lead to the apprehension of the suspects, to urgently contact police via the Crime Stop number 08600 10111 or the My SAPS app that can be downloaded onto any smartphone."
